RESUMO

The objective of this study was to describe nutritional strategies utilized on Canadian dairy farms with automated milking systems (AMS), both at the feed bunk and the concentrate offered at the AMS, as well as to determine what dietary components and nutrients, as formulated, were associated with milk production and milking behaviors on those farms. Formulated diets, including ingredients and nutrient content, and AMS data were collected from April 1, 2019, until September 30, 2020, on 160 AMS farms (Eastern Canada [East] = 8, Ontario [ON] = 76, Quebec [QC] = 22, and Western Canada [West] = 54). Both partial mixed ration (PMR) and AMS concentrate samples were collected from May 1 to September 30, 2019, on 169 farms (East = 12, ON = 63, QC = 42, West = 52). AMS milking data were collected for 154 herds. For each farm (n = 160), milk recording data were collected and summarized by farm to calculate average milk yield and components. Multivariable regression models were used to associate herd-level formulated nutrient composition and feeding management practices with milk production and milking behavior. Milk yield (37.0 ± 0.3 kg/d) was positively associated with the PMR ether extract (EE) concentration (PMR % EE; +0.97 kg/d per percentage point (p.p.) increase) and with farms that fed barley silage as their major forage source on farm (n = 16; +2.18 kg/d) compared with haylage (n = 42), while farms that fed corn silage (n = 96; +1.23 kg/d) tended to produce more milk than farms that fed haylage. Greater milk fat content (4.09 ± 0.28%) was associated with greater PMR-to-AMS concentrate ratio (+0.02 p.p. per unit increase) and total diet net energy for lactation (+0.046 p.p. per 0.1 Mcal/kg increase), but lesser % non-fiber carbohydrates (NFC) of the PMR (-0.016 p.p. per p.p. increase of % NFC). Milk protein content (3.38 ± 0.14%) was positively associated with forage % of the PMR (+0.003 p.p. per p.p. increase of % forage) and total diet % starch (+0.009 p.p. per p.p. increase of % starch), but negatively associated with farms feeding corn silage (-0.1 p.p. compared with haylage) as their major forage. Greater milking frequency (2.77 ± 0.40 milkings/d) was observed on farms with free-flow cow traffic systems (+0.62 milkings/d) and positively associated with feed push-up frequency (+0.013 milkings/d per additional feed push-up), while being negatively associated with PMR NFC content and % forage of the total ration (-0.017 milkings/d per p.p. increase of % forage). Lastly, greater milking refusal frequency (1.49 ± 0.82 refusals/d) was observed on farms with free-flow cow traffic systems (+0.84 refusals/d) and farms feeding barley silage (+0.58 refusals/d) than guided flow and farms feeding either corn silage or haylage, respectively. These data give insight into the ingredients, nutrient formulations and type of diets fed on AMS dairy farms across Canada and the association of those factors with milk production and milking behaviors.

The objective of this study was to determine the effect of individual cow personality traits on feeding behavior and production under low levels of feeding competition, and to determine whether personality traits influence how feeding behavior changes in response to greater feeding competition. Forty-two Holstein cows were assigned to 1 automated feed bin per cow (low competition condition) from 15 to 28 d in milk (DIM; period 1, P1), and 2 feed bins per 3 cows (higher competition condition) from 63 to 76 DIM (period 2, P2). A total mixed ration (TMR) was fed into the automated feed bins which recorded each feed bin visit time, duration, and intake. Cow personality traits were assessed at 21 DIM during P1 and at 70 DIM during P2 using a combined arena test, measuring behavioral responses to a novel environment, novel object, and novel human. Principal components analysis of behaviors observed during the P1 combined arena test revealed 1 factor (interpreted as active-explorative) from the novel environment test explaining 51% of the variance, and 3 factors (interpreted as fearfulness, active-explorative, and sociability toward conspecifics) from each of the novel object (76% cumulative variance) and human (75% cumulative variance) tests. The principal components analysis of behaviors observed during the P2 combined arena test revealed 2 factors jointly from the environment, object, and human tests (interpreted as fearfulness and active-explorative) that together explained 68% of the variance. Fearfulness and active-explorative trait scores were correlated across P1 and P2, indicating stability of personality over a challenging period and advancing DIM. In P2 when competition for feed was increased at greater stage of lactation, the more active-explorative cows appeared to make few alterations to their feeding behavior, yet still maintained their milk yield, compared with lower competition in P1. In contrast, cows who were more fearful increased their feed bin visits from P1 to P2, and less fearful cows increased their eating rate, without increased milk production, despite advanced lactation. Overall, the results indicate that cows of different personalities adopt different feeding strategies in response to a change in their environment, and may benefit from tailored management during challenging periods.

The objective of this study was to quantify the effects of supplementing a low level of dry glycerol product pre- and postpartum on the feeding behavior, lying behavior, and reticulorumen pH of dairy cows. Multiparous Holstein dairy cows (n = 60) were enrolled in a 2 × 2 factorial design study. Twenty-one days before expected parturition, cows individually received a dry cow diet with (1) 250 g/d glycerol supplementation (GLY; 66% pure glycerol, United States Pharmacopeia grade), or (2) no supplementation (CON). Following parturition, cows were individually assigned to either (1) 250 g/d glycerol product (GLY; 66% pure glycerol), or (2) no supplementation (CON) to their partial mixed ration (PMR) for the first 21 d in milk (DIM). All cows were milked by an automated milking system and offered a target of 5.4 kg/d pellet (23% of target total dry matter intake [DMI]). For both treatment periods, cows were individually assigned to automated feed bins to measure PMR feeding behavior. Rumination time and lying behavior were monitored with electronic sensors for the whole study (-21 to 21 DIM). Reticulorumen pH boluses were administered to a subset of cows (n = 40) where pH was recorded every 10 min from 21 d prepartum to 21 d postpartum. Prepartum, cows fed GLY had fewer, larger meals and spent 20.2% more time feeding than CON while consuming feed at a similar rate. Cows on the CON diet prepartum spent more time lying down in more frequent bouts in the 21 d before calving. Following parturition, cows that received GLY prepartum continued to devote more time to eating, while tending to spend less time ruminating per kilogram of DMI. Cows receiving CON postpartum had larger meals with longer intervals between meals. In the first 21 DIM, cows receiving CON prepartum tended to have shorter, but significantly more frequent, lying bouts than cows fed GLY prepartum. Glycerol supplementation pre- and postpartum resulted in less time spent lying down following parturition. Minimal differences between treatments were observed for pre- and postpartum sorting behavior or reticulorumen pH. Overall, supplementation of glycerol pre- and postpartum altered cow time budgets, with cows spending more time eating pre- and postpartum, less time lying pre- and postpartum, and having fewer, larger meals prepartum when receiving glycerol prepartum, and with cows having slower feeding rates and smaller meals following parturition with postpartum glycerol supplementation.

This observational study aimed to determine the association of cow-level factors and herd-level housing and management practices during the dry period with indicators of udder health in early-lactation cows in automated milking system (AMS) herds. Data were collected from 166 commercial AMS dairy farms (mean ± standard deviation = 116 ± 111 milking cows; range = 39 to 1,200) across Canada between October 2018 and September 2020. Information on herd demographics, housing, and management practices was obtained on each farm using 2 surveys. On each farm, we selected all cows that had available Dairy Herd Improvement (DHI) somatic cell count (SCC) data for their last milk test before dry-off (>250 d in milk) and their first milk test after calving (5-45 d in milk). Data from 14,007 cows were included after excluding cows with a dry period of <30 d and >120 d. Using the SCC data, we calculated for each cow the somatic cell score (SCS) for the last milk test before dry-off (PreSCS) and the first milk test after calving (PostSCS), which we then averaged per herd at a test-day level. Intramammary infection (IMI) was estimated using cow SCC data. Each cow was classified as not infected (SCC <200,000 cells/mL) or infected (SCC ≥200,000 cells/mL) at her last milk test before dry-off and her first milk test after calving. Based on this classification, cows were further categorized as never infected, always infected, new IMI, or cured IMI. At the cow level, a higher PostSCS was associated with longer dry periods. The odds of having a new IMI were higher for cows of higher parity and that had lower 305-d milk yield before dry-off. Cows with lower parity were more likely to cure an IMI. At the herd level, a higher 305-d milk yield before dry-off was associated with a lower incidence of new IMI and a higher incidence of cured IMI. Separating cows into a different pen as preparation for dry-off tended to be associated with a lower PostSCS and incidence of new IMI. At dry-off, herds that used teat sealants and blanket antibiotic dry cow therapy also had lower PostSCS. During the dry period, housing cows in different groups was associated with a higher PostSCS and a lower incidence of cured IMI, while housing cows in both pack pens and stalls compared with only pack pens was associated with a lower incidence of new IMI. Finally, placing cows onto the AMS to be milked one or more days after calving tended to be associated with a lower PostSCS compared with placing them in the AMS within the first day postpartum. In summary, indicators of udder health in early-lactation cows in AMS herds were associated with several cow-level factors and herd-level housing and management practices before dry-off, at dry-off, during the dry period, and at the beginning of lactation. Thus, if some of the associations identified are causal, AMS producers may be able to improve udder health through modifications of housing and management practices.

Lameness and leg injuries are both painful and prevalent across the dairy industry, and are a major welfare concern. There has been a considerable amount of research focused on investigating the risk factors associated with lameness and injuries and how they might be prevented and treated. The objectives of this narrative review were to summarize herd-level prevalence estimates, risk factors, strategies for prevention, control, and treatment of these conditions, and the barriers to best practice adoption for lameness and injuries on dairy farms. There is a relatively high within-herd prevalence of lameness on dairy farms globally, with a recent systematic review estimating the mean prevalence at 22.8%. Similarly, there is a relatively high prevalence of hock injuries, with within-herd estimates ranging from 12 to 81% of cows affected. Knee and neck injuries have been reported to be less common; 6-43% and 1-33%, respectively. Numerous risk factors have been associated with the incidence of lameness, notably housing (e.g., access to pasture, bedding depth, bedding type, flooring type, stall design), management (e.g., stall cleanliness, frequency of trimming, holding times, stocking density), and cow-level (e.g., body condition, parity, injured hocks) factors. Risk factors associated with hock injuries can be similarly classified into housing (e.g., bedding type and depth, outdoor access, parlor type, stall design), management (e.g., bedding depth, cleanliness), and cow (e.g., parity, days in milk, lameness) factors. Key preventative approaches for lameness include routine preventative and corrective hoof trimming, improving hoof cushioning and traction through access to pasture or adding rubber flooring, deep-bedded stalls, sand bedding, ensuring appropriate stocking densities, reduced holding times, and the frequent use of routine footbaths. Very little research has been conducted on hock, knee, and neck injury prevention and recovery. Numerous researchers have concluded that both extrinsic (e.g., time, money, space) and intrinsic (e.g., farmer attitude, perception, priorities, and mindset) barriers exist to addressing lameness and injuries on dairy farms. There are many diverse stakeholders in lameness and injury management including the farmer, farm staff, veterinarian, hoof trimmer, nutritionist, and other advisors. Addressing dairy cattle lameness and injuries must, therefore, consider the people involved, as it is these people who are influencing and implementing on-farm decisions related to lameness prevention, treatment, and control.

This study aimed to determine if personality traits identified in dairy cows during the transition phase are correlated with those of their peripubertal heifer calves. At ~24 d before calving and ~24 d after being first introduced to the automated milking system, the personality traits of 23 Holstein cows were assessed using a combined arena test [consisting of exposure to consecutive novel environment (NE), novel object (NO), and novel human (NH) tests]. Personality traits were established by principal component analysis (PCA) of behaviors expressed in these tests. The PCA of cow behaviors during the precalving test revealed 3 factors, or personality traits, interpreted as exploratory, active, and bold. The PCA of cow behaviors during the postcalving test revealed 2 factors, interpreted as active and exploratory. From these cows, 23 female Holstein heifers were produced and enrolled in this study at 7 mo of age. Heifers were personality tested once through a similar combined arena test. The PCA of heifer behaviors during the NO test revealed 3 factors, interpreted as bold, exploratory-active, and social. The PCA of heifer behaviors during the NH test revealed 2 factors, interpreted as exploratory-active and social. All factor scores from each cow and heifer pair were tested for association. An association between cows with higher scores on the factor interpreted as exploratory and heifers with lower scores on the factor interpreted as bold was detected. There were tendencies for cows with higher scores on the factor interpreted as active to be associated with heifers that scored highly on the factors interpreted as exploratory-active and bold. The data suggests that there are some limited associations between personality traits of cows and their heifer offspring; further exploration of these associations may lead to the prediction of heifer personality based on cow personality.

The objective of this study was to quantify the effects of supplementing transition dairy cows with a low inclusion dry glycerol product in the pre- and postpartum periods on feed intake, metabolic markers, and milk yield and components. Multiparous Holstein dairy cows (n = 60) were enrolled in a 2-by-2 factorial design study. Starting 21 d before expected parturition, cows individually received a dry cow diet with (1) 250 g/d glycerol product supplementation [66% pure glycerol (United States Pharmacopeia grade); GLY], or (2) no supplementation (CON) mixed to their total mixed ration. After parturition, cows, again, were individually assigned to either GLY, or (2) no supplementation (CON) to their partial mixed ration for the first 21 d in milk (DIM). Cows were milked by an automated milking system and offered a target of 5.4 kg DM/d pellet (23% of target total dry matter intake, DMI) in the automated milking system and followed for 42 d into lactation. Blood samples were collected 6.3 ± 3.47 d before calving for all blood measures and 3, 7, 10, and 14 DIM for analysis of glucose and ß-hydroxybutyrate, as well as 3 and 7 DIM for nonesterified fatty acids (NEFA) and haptoglobin. Initial dry cow body weight (BW), calf birth weight, previous 305-d milk, and month of parturition were used as covariates in the statistical model. Cows supplemented with GLY prepartum lost less BW and consumed more DMI pre- and postpartum, as well as had lower postpartum blood ß-hydroxybutyrate and NEFA concentrations compared with those fed the CON treatment prepartum. Cows supplemented with GLY postpartum had lesser DMI in the first 42 DIM than cows fed CON postpartum, but also had reduced blood NEFA concentrations, odds of a high haptoglobin test, odds of a low blood glucose test, and lesser preformed fatty acid concentrations and yields in their milk. Cows supplemented glycerol both pre- and postpartum lost the least total BW from -21 to 21 DIM. No treatment effects were detected for milk yield; however, cows receiving GLY postpartum had lower milk fat. Overall, glycerol supplementation during the transition period, particularly during the 21 d before calving, was associated with markers of improved metabolic status.

Studies evaluating the crude protein (CP) supplementation strategies across the year for grazing cattle and its association with the enzymes involved in the urea cycle and muscle and mammary gland developments are scarce. Thus, we aimed to evaluate the effect of supplementation with different levels of CP on the expression of genes involved in the urea cycle and muscle and mammary gland development of Holstein × Gyr crossbreed heifers grazing intensively managed Brachiaria decumbens throughout the year. Thirty-eight heifers with average initial BW of 172.5 ± 11.15 kg (mean ± SE) and 8.2 ± 0.54 mo of age were randomly assigned to 1 of 4 treatments: 3 protein supplements (SUP) fed at 5g/kg of body weight, plus a control group (CON, non-supplemented animals). The supplement CP levels evaluated were: 12, 24, and 36%. The study was divided into 4 seasons: rainy, dry, rainy-dry transition (RDT), and dry-rainy transition (DRT). On the penultimate day of each season, ultrasound images of the carcass and mammary gland were taken. Five animals from each treatment were randomly chosen on the last day of each season, and liver and muscle tissue biopsies were performed. The target genes were the mammalian target of rapamycin (mTOR) and adenosine monophosphate-activated protein kinase (AMPK) in the muscle samples. Carbamoyl phosphate synthetase (CPS), ornithine transcarbamylase (OTC), argininosuccinate synthetase (ASS), arginosuccinate lyase (ASL), and arginase (ARG) were evaluated in the liver samples. Data were analyzed using PROC GLIMMIX of the SAS with repeated measures. We observed a greater rib eye area (cm2) and fat thickness (mm) in SUP animals than in non-supplemented animals. However, we did not observe differences among SUP levels for both variables. No effects of supplementation were detected on mammary gland development. Nevertheless, seasonal effects were observed, where the RDT and dry season had the most and least accumulated fat in the mammary gland. In muscle, we observed greater expression of AMPK in non-supplemented animals than SUP animals. On the other hand, no differences were observed in gene expression between SUP and non-supplemented animals and among SUP animals for mTOR. Season affected both AMPK and mTOR; heifers had a greater AMPK gene expression on rainy than RDT. For mTOR, we observed greater gene expression in RDT and DRT than in rainy. No differences were observed among RDT, dry, and DRT, and between dry and rainy seasons for mTOR. We observed greater CPS, ASL, and ARG gene expression in SUP animals than in non-supplemented animals. Among SUP animals, supplement CP linearly affected CPS. In conclusion, the supplementation strategy did not affect mammary gland development and mTOR expression in muscle tissue. However, we observed a seasonal effect on mammary gland development and AMPK and mTOR expression. The CP supplementation increased the rib eye area and fat thickness, directly affecting AMPK expression in the muscle. Moreover, the CP supplementation increased urea cycle enzyme expression, indicating greater urea production in the liver.

Echinacea purpurea (EP) is an herb that has demonstrated immunostimulatory and anti-inflammatory effects with the potential to improve immunity, health, and performance in animals. The objective of this study was to investigate how supplementing calves with EP affects their blood immunity marker profile, health, intake, and growth. Male Holstein calves (n = 240), sourced from local dairy farms or auction, arrived at a rearing facility between 5 and 14 d of age and were kept in individual pens in 1 of 3 rooms (80/room) for 56 d, and then put into groups for the remaining 21 d of the trial. Calves received milk replacer (MR) 2× per day for 56 d (total = 36 kg of MR) and had ab libitum water and starter access. Within room, calves were randomly assigned to 1 of 3 treatments: (1) control (n = 80), (2) 3g of dried (powder) EP extract per day split over 2 milk feedings from experiment d 14-28 (n = 80), and (3) 3 g of dried (powder) EP extract per day split over 2 milk feedings from experiment d 1-56 (E56; n = 80). The powdered EP treatments were mixed into the liquid MR. On d 1, 14, 28, and 57 rectal temperatures and blood were collected from a subset of calves (n = 117; 39 calves/treatment), and blood serum was assessed for serum total protein (d 1), haptoglobin, white blood cells, and cytokines. Failed transfer of passive immunity was defined as serum total protein <5.2 g/dL. Calves were health scored 2× per day, receiving fecal and respiratory scores until d 28 and 77, respectively. Calves were weighed on arrival and then weekly until d 77. Milk replacer and feed refusals were recorded. Supplementation of EP was associated with lower haptoglobin levels, segmented neutrophil counts, segmented neutrophil per lymphocyte ratio, respiratory scores in auction derived calves, and higher lymphocyte counts and d 28 rectal temperature. Of calves with heavier arrival body weight, E56 calves had greater postweaning weekly body weight. There was no detected effect of EP supplementation on total white blood cells, band neutrophil, monocyte, and basophil counts, IL-10, IL-6, and TNF-α levels, fecal scores, risk of receiving diarrhea and respiratory treatment, risk of bovine respiratory disease (calves were deemed at risk for bovine respiratory disease if they had at least 1 respiratory score ≥5), risk of mortality, MR and feed intake, average daily gain, and feed conversion ratio. Overall, EP supplementation to dairy calves was associated with immunomodulation and reduced inflammation, evidenced through blood markers, although only few minor health and growth improvements were observed. Benefits were observed particularly when fed across the whole milk feeding period.

The objectives of this study were to determine how dairy cow personality traits affect their adaptation to an automated milking system (AMS) upon parturition, as well as whether these personality traits are consistent across the transition from gestation to lactation. Sixty Holstein dairy cows (19 primiparous and 41 multiparous) were assessed for personality traits using a combined arena test conducted at 24 d before parturition and 24 d after first introduction to an AMS, which occurred ∼3 d after parturition. The combined arena test comprised 3 parts: a novel arena test, a novel object test, and a novel human test. Principal component analysis of the behaviors recorded during the personality assessment revealed 3 factors interpreted as personality traits (75% cumulative variance) in the pre-calving test, interpreted as explore, active, and bold. The post-calving test revealed 2 factors (78% cumulative variance), interpreted as active and explore. Data from d 1 to 7 after introduction to the AMS were summarized by cow and associated with the pre-calving factors, and data from d 21 to 27 after introduction to the AMS were summarized by cow and associated with the post-calving factors. The active trait had a moderate positive correlation between the pre- and post-calving tests, whereas exploration had a weak positive correlation between tests. Cows that scored high for activeness in the pre-calving test tended to have fewer fetching events and a higher coefficient of variation of milk yield in the first 7 d after introduction to the AMS, whereas bolder cows tended to have higher milk yield during that period. In the post-calving test, more active cows tended to have more frequent milkings and voluntary visits per day, as well as a lower cumulative milk yield from d 21 to 27 after introduction to the AMS. Overall, these results indicate that personality traits of dairy cows are associated with adaptation and performance in an AMS, and that personality traits are consistent across the transition period. Specifically, cows that scored high for boldness and activeness adapted better to the AMS immediately after calving, whereas cows that scored low for activeness and high for boldness performed better in terms of milk yield and milking activity in early lactation. This study demonstrates that personality traits affect milking activity and milk yield of dairy cows milked with an AMS and, therefore, may be useful for selection of cows who might best adapt to and use an AMS.

The objective of this study was to determine whether associations exist between position while ruminating (lying vs. standing) and milk and component production in dairy cows. Data from 30 lactating Holstein cows were assembled from 2 studies in which cows were milked by automated milking system (AMS) and fed a partial mixed ration (PMR) in feed bins that recorded intake behavior. Rumination and lying behavior were monitored using automated neck- and leg-based sensors, respectively. Each cow was monitored over 2 separate 2-wk treatment periods. To estimate position while ruminating for each 2-h period of the day for each cow, a conditional probability was calculated to determine the probability that any rumination time and lying time were occurring at the same time in any 2-h period. These probabilities (RwL), and all behavioral data, were summarized per cow per 2-h interval, and then averaged per day and per 2-wk period, along with milk yield and component data. Cows averaged (mean ± standard deviation) 1.9 ± 1.1 lactations and 85.5 ± 55.2 d in milk, and weighed 668.5 ± 96.0 kg. Data included rumination time (557.7 ± 41.1 min/d), lying time (703.9 ± 65.3 min/d), idle standing time (520.1 ± 83.2 min/d), PMR feeding time (204.7 ± 48.5 min/d), PMR dry matter intake (DMI; 21.8 ± 4.6 kg/d), AMS pellet provision (4.6 ± 1.6 kg/d), total DMI (26.4 ± 4.5 kg/d), milk yield (42.4 ± 7.2 kg/d), milk fat content (3.75 ± 0.51%), and milk protein content (3.21 ± 0.32%). Greater rumination time and lying time were associated with greater RwL probability (mean = 0.19 ± 0.02; range = 0.14 to 0.23). The RwL probability tended to be positively associated with total DMI and milk fat content, was associated with milk protein content, but was not associated with any measures of milk yield. The results indicate that in a free-traffic AMS, cows who have greater probability of ruminating while lying down spend more time ruminating and lying, and tend to consume more total dry matter and produce milk with greater components.

Ketosis in dairy cattle is primarily diagnosed based on the concentrations of ketone bodies in the blood, milk, or urine. Cow-side tests using these fluids are available for rapid detection of elevated concentrations of ketone bodies. Although these tests have been extensively validated, the performance of different tests has not been compared over time. Our objectives were to investigate the relationship between point-of-care diagnostic tests measuring the concentrations of ß-hydroxybutyrate (BHB) in blood (BT; Precision Xtra, Abbott Laboratories), BHB in milk (MT; Keto-Test, Elanco), and acetoacetate (AcAc) in urine (UT; Ketostix, Bayer Corporation) through cases of ketosis. Holstein cows (n = 148) were screened daily for hyperketonemia (HYK; blood BHB ≥1.2 mmol/L) from 3 to 16 d in milk (DIM); moreover, milk and urine samples were collected concomitantly and tested for ketones (ketosis thresholds: 100 µmol/L milk BHB and 5 mg/dL urine AcAc). Of the animals screened (n = 148), 74% were diagnosed with HYK. When diagnosed with HYK, cows were treated with propylene glycol orally once daily for 5 d. After the day of diagnosis (d 0), hyperketonemic cows were retested with BT, MT, and UT for 3 d (d 1, 2, and 3). We assessed the diagnostic test performance and time to ketosis (survival analyses and Cox proportional hazards models) of MT and UT compared with BT. Considering all paired samples (before and after diagnosis of HYK), MT had 61% sensitivity and 91% specificity, whereas the UT had 77% sensitivity and 94% specificity compared with BT. The specificity of MT and UT increased from d 0 to d 1, decreased on d 2, and increased on d 3. The median time to diagnosis of ketosis in blood was 5 DIM (95% CI 5 to 7 DIM); moreover, MT and UT had 2 d greater median time to diagnosis of ketosis compared with the BT [7 DIM (6 to 11 d); and 7 DIM (6 to 13 d), respectively]. We concluded that the UT is a more sensitive predictor of blood BHB concentration than the MT. The UT and MT tests diagnosed ketotic cows approximately 2 d later than the BT. The possible consequences of delay in detection of ketosis in milk and urine should be investigated.

Our objectives were to evaluate the effects of complete replacement of supplementary inorganic salts of trace minerals (STM) by organic trace minerals (OTM) in both pre- and postpartum diets on feeding behavior, ruminal fermentation, rumination activity, energy metabolism, and lactation performance in dairy cows. Pregnant cows and heifers (n = 273) were blocked by parity and body condition score and randomly assigned to either STM or OTM diets at 45 ± 3 d before their expected calving date. Both groups received the same diet, except for the source of trace minerals (TM). The STM group was supplemented with Co, Cu, Mn, and Zn sulfates and Na selenite, whereas the OTM group was supplemented with Co, Cu, Mn, and Zn proteinates and selenized yeast. Treatments continued until 156 days in milk and pre- and postpartum diets were formulated to meet 100% of recommended levels of each TM in both treatments, taking into consideration both basal and supplemental levels. Automatic feed bins were used to assign treatments to individual cows and to measure feed intake and feeding behavior. Rumination activity was monitored by sensors attached to a collar from wk -3 to 3 relative to calving. Blood metabolites were evaluated on d -21, -10, -3, 0, 3, 7, 10, 14, 23, and 65 relative to calving. Ruminal fluid samples were collected using an ororuminal sampling device on d -21, 23, and 65 relative to calving, for measurement of ruminal pH and concentration of volatile fatty acids. Cows were milked twice a day and milk components were measured monthly. Cows supplemented with OTM tended to have longer daily feeding time (188 vs. 197 min/d), and greater dry matter intake (DMI; 12.9 vs. 13.3 kg), and had a more positive energy balance (3.6 vs. 4.2 Mcal/d) and shorter rumination time per kg of dry matter (DM; 40.1 vs. 37.5 min/kg of DM) than cows supplemented with STM during the prepartum period. In the postpartum period, OTM increased DMI in multiparous cows (24.1 vs. 24.7 kg/d) but not in primiparous cows (19.1 vs. 18.7 kg/d). The difference in DMI of multiparous cows was more evident in the first 5 wk of lactation, when it averaged 1 kg/d. Milk yield was not affected by treatment in multiparous cows (44.1 vs. 44.2 kg/d); however, primiparous cows supplemented with OTM had lesser yields than primiparous cows supplemented with STM (31.9 vs. 29.8 kg/d). Cows supplemented with OTM had a greater percentage of protein in milk (3.11 vs. 3.17%), reduced concentration of nonesterified fatty acids in serum (0.45 vs. 0.40 mmol/L), and rumination activity (30.1 vs. 27.8 min/kg of DM) than cows supplemented with STM. At the end of the transition period, cows supplemented with OTM had reduced molar proportion of acetate, reduced pH, and tended to have a greater concentration of total volatile fatty acids in ruminal fluid. In conclusion, complete replacement of STM by OTM caused modest changes in rumen fermentation, feeding behavior, energy metabolism, and performance of dairy cows, improving postpartum DMI in multiparous cows and reducing circulating levels of nonesterified fatty acids. The pre-absorptive effects of TM source and the parity specific responses on performance warrant further research.

This study was designed to develop a protocol for repeated intramuscular indomethacin injections to replicate leaky-gut-like symptoms in male Holstein calves to model and study the detrimental effects of leaky gut on gut tissue function and inflammatory response. A generalized randomized block design was used to evaluate how repeated indomethacin intramuscular injections affected the development of leaky gut in 18 male Holstein calves. Animals were enrolled at 3 ± 1 d of life, and after 21 d of adaptation, they were randomly assigned to 1 of 3 treatments consisting of intramuscular saline or indomethacin injections every 12 h for 48 h: (1) control (CTL), saline injection, (2) low intramuscular indomethacin (INDO-L) dosed at 1.2 mg/kg of body weight (BW), and (3) high intramuscular indomethacin (INDO-H) dosed at 2.4 mg/kg of BW. During the challenge, milk intake, starter intake, fecal scores, and rectal temperature were measured daily, and BW was measured at the beginning and at the end of the challenge. Plasma samples were used to measure the recovery of markers of intestinal permeability before and after the challenge by dosing lactulose, d-mannitol, and chromium-EDTA. In addition, several cytokines were measured in plasma during the challenge. Calves were dissected at the end of the challenge to obtain tissue and digesta samples from the gastrointestinal tract and liver. No treatment differences were observed for starter and milk intakes, fecal scores, BW, and rectal temperature. The difference in marker concentrations between pre and post challenges was higher for INDO calves compared with CTL calves in the case of lactulose and chromium-EDTA. In addition, chemokine ligand 2 and 4 and IL-6 were higher for INDO-H calves compared with CTL. Both doses of indomethacin resulted in reductions in villus length and surface area in the distal jejunum and ileum and reductions in crypt depth and width in the colon. We showed that repeated indomethacin injections over a 48-h period induced leaky-gut-like symptoms in a region-specific manner, affecting mainly the distal section of the intestine. This outcome was characterized by histomorphological changes in the distal jejunum, ileum, and colon and by increased gut permeability. Interestingly, changes in liver morphology and immune function also occurred, possibly due to the increased translocation of foreign antigens breaching the epithelial cell wall. The leaky gut challenge model described here could be used to improve understanding of the pathogenesis of intestinal disorders in cattle and provide a reliable alternative for testing feed additives with intestinal health benefits.

The primary objective of this study was to determine whether the level of concentrate allowance in an automated milking system (AMS) affects the feed intake, eating behavior, milking activity, and performance of lactating dairy cows. The secondary objective of this study was to describe how the response to concentrate allocation, specifically in feeding and milking behavior, varies with cow personality traits. Fifteen Holstein cows were used in a crossover design with two 28-d periods, each including 14 d of adaptation and 14 d of data collection. The cows were housed in a freestall pen with free-traffic access to the AMS. Treatments consisted of a basal partial mixed ration (PMR) common to both treatment groups, with a concentrate allowance (on dry matter basis) of (1) 3.0 kg/d in the AMS (L-AMS) or (2) 6.0 kg/d in the AMS (H-AMS). Between the 2 treatment periods, each cow was assessed for personality traits using a combined arena test consisting of exposure to a novel environment, novel object, and novel human. Principal component analysis of behaviors observed during the novel environment and object tests revealed 3 factors (interpreted as active, social, and alert-curious) that together explained 76% of the variance, whereas principal component analysis of the novel human test revealed 2 factors (interpreted as active-vocal and fearful of novel humans) that together explained 77% of the variance. When on the H-AMS treatment, PMR dry matter intake (DMI) was less (24.5 vs. 26.0 kg/d) and AMS concentrate delivery was greater (5.9 vs. 3.1 kg/d), as per design. Consequently, total DMI was greater on the H-AMS treatment (30.4 vs. 29.1 kg/d). When on the H-AMS treatment, cows who were more alert-curious consumed more PMR, whereas cows who were more fearful of the novel human were less likely to receive the maximum amount of AMS concentrate available, limiting their total DMI and increasing the day-to-day variability of that intake. Although this was a preliminary study, these data suggest an association between dairy cow personality traits and how cows respond to increased AMS concentrate allowance.

Milk fatty acid (FA) profile can be divided into (1) de novo (C4-C14) that are synthesized in the mammary gland; (2) preformed (≥C18) that are absorbed from blood and originate from mobilized adipose tissues or dietary fat; and (3) mixed (C16), which have both origins. Our objectives were to describe the FA profile, as predicted using Fourier transform mid-infrared spectroscopy, of bulk tank milk from automated milking system (AMS) farms and to assess the association of management and housing factors with the bulk tank milk composition and FA profile of those AMS farms. The data used were collected from 124 commercial Canadian Holstein dairy farms with AMS, located in the provinces of Ontario (n = 68) and Quebec (n = 56). The farms were visited once from April to September 2019, and information were collected on barn design and herd management practices. Information regarding individual cow milk yield (kg/d), days in milk, parity, and the number of milking cows were automatically collected by the AMS units on each farm. These data were extracted for the entire period that the bulk tank milk samples were monitored, from April 2019 to April 2020 in Quebec and from August 2019 to May 2020 in Ontario. Across herds, milk yield averaged (mean ± standard error) 35.9 ± 0.4 kg/d, with 3.97 ± 0.01% fat and 3.09 ± 0.01% protein, whereas FA profile averaged 26.2 ± 0.1, 33.1 ± 0.1, and 40.7 ± 0.2 g/100 g of FA for de novo, mixed, and preformed, respectively. The FA yield averaged 0.34 ± 0.01, 0.44 ± 0.01, and 0.54 ± 0.01 kg/d for de novo, mixed, and preformed, respectively. Multivariable regression models were used to associate herd-level housing factors and management practices with milk production, composition, and FA profile. Milk yield was positively associated with using a robot feed pusher (+2.1 kg/d) and the use of deep bedding (+2.6 kg/d). The use of a robot feed pusher, deep bedding, and greater stall raking frequency were positively associated with greater yield (kg/d) of de novo, mixed, preformed, and de novo + mixed FA. Use of deep bedding was negatively associated with concentration of fat, de novo FA, mixed FA, and de novo + mixed FA, expressed in grams per 100 g (%) of milk. A wider lying alley width (≥305 cm) was associated with a greater concentration (g/100 g of milk) of de novo and de novo + mixed FA. Greater frequency of partial mixed ration delivery (>2×/d vs. 1 and 2×/d) was positively associated with a greater proportion (g/100 g of FA) of de novo, mixed, and de novo + mixed FA and negatively associated with the proportion of preformed FA. Overall, these associations indicated that bulk tank FA profile can be used as a tool to monitor and adjust management and housing in AMS farms.

Milk production may be reduced before dry-off to decrease the risk of cows developing intramammary infections during the dry period. Such reductions in milk may be possible in automated milking systems (AMS) where milking frequency and feed allocation at the AMS can be controlled at the cow level. This study investigated the effect of dry-off management of cows milked in AMS on milk yield, milking behavior, and somatic cell count (SCC). Using a 2 × 2 factorial arrangement of treatments, applied from d 14 to 1 before dry-off, 445 cows from 5 commercial dairy farms in Quebec, Canada, were assigned within farm to either (1) reduced feed [RF; allowed a maximum of 0.75 kg/d of AMS pellet for the first week (14 to 8 d before dry-off) of treatment, and 0.50 kg/d for the second week (7 to 1 d before dry-off) of treatment], or (2) nonreduced feed (NF; allowed up to 2 kg/d of AMS pellet), and either (1) reduced milking (RM; reduced to 2 milkings/d or as many times as required to yield 17 kg/milking), or (2) nonreduced milking (NM; allowed up to 6 AMS milkings/d) and no maximum production. Feed and milking behavior data, as well as milk yield and SCC were collected from the AMS software. The RF cows had lower AMS feed delivered during the treatment period, as per the experimental design. Across the treatment period, the NF-NM cows had the highest milking frequency (2.7 times/d), followed by the RF-NM cows (2.4 times/d), and then both of the RM groups (1.8 times/d), which did not differ from each other. All cows, except the NF-NM cows, were gradually milked less frequently as dry-off approached. Across the entire 2-wk treatment period before dry-off, cows with RM allowance experienced a higher reduction in milk yield compared with the cows with no milking allowance restrictions (-4.8 vs. -3.6 kg). Similarly, cows with a RF allocation tended to have a higher reduction in milk yield than cows with NF (-4.6 vs. -3.7 kg). As result, those cows with both reduced milking permissions and feed allocation at the AMS experienced the greatest drop in milk production before dry-off. There were no differences between treatments for milking frequency or yield in the next lactation. Somatic cell score (calculated from SCC) was not different between treatments in the 2-wk or day before dry-off, nor in the first month after calving. Overall, these data suggest that reducing both milking frequency and feed quantity in the AMS is the most efficient method to decrease milk yield before dry-off, without negatively influencing milking frequency or yield in the next lactation, as well as without affecting milk quality.

The objective of this study was to investigate the effects of a milk byproduct-based starter pellet, high in lactose, on feed intakes, growth, and rumen development when calves were fed either a high or low level of milk. This was done by using a 2 × 2 factorial design, with 120 Holstein heifer calves randomly assigned to 1 of 2 milk treatments and 1 of 2 feed treatments upon enrollment at birth. Calves were either assigned to a low (LM) or high (HM) level of milk replacer by an automated milk-feeding system for individually-housed calves. The LM treatment calves were offered 6 L/d (150 g/L; 1.2 L, 5×/d), whereas HM treatment calves were offered 15 L/d (150 g/L; 3 L, 5×/d) of milk replacer. Calves were also assigned to a milk byproduct- (whey) based starter feed treatment (WF), or a grain-based starter ration (95% concentrate starter pellet, 5% chopped wheat straw) feed treatment (GF). All calves were offered their treatment feed starting on d 5, with water offered ad libitum from d 0. The WF treatment calves were offered 150 g/d of the milk byproduct-based pellet until they consumed the entirety for 2 out of 3 consecutive days. At this point, WF treatment calves received 150 g of the milk byproduct-based pellet top dressed onto the grain-based ration to allow for ad libitum feed consumption until weaned, when they only received the grain-based ration postweaning (d 64-77). Calves on the GF feed treatment received only the grain-based ration. All calves were gradually weaned from d 43 to 63. By design, calves on the HM treatment consumed greater levels of milk than LM treatment calves for the majority of the days in the preweaning phase (8.9 ± 0.11 vs. 5.5 ± 0.11 L/d; SE). Calves on the GF and WF treatments consumed similar amounts of milk in the preweaning phase (7.2 ± 0.11 vs. 7.2 ± 0.11 L/d); however, there was a milk and feed treatment interaction detected for milk consumption in the weaning phase, with HM-GF calves drinking more milk than the other treatments and HM-WF calves drinking more milk than both LM-GF and LM-WF calves. The WF treatment calves had greater solid feed intake during the preweaning phase than GF treatment calves, with no feed intake differences in the weaning or postweaning phases. The WF calves drank more water in the preweaning phase than GF calves. Average daily gain was similar throughout the study between the GF and WF treatment calves. Rumen physiological measures were all similar by the end of the trial (d 77) regardless of feed or milk treatment, suggesting similar rumen development postweaning. Overall, regardless of the level of milk fed, offering the milk byproduct-based starter pellet may increase preweaning feed intake, along with water consumption. However, both feed and milk treatments had similar performance and rumen development measures by the end of the trial.

This randomized controlled trial investigated the effects of temporarily reducing milking frequency (MF) on the resolution of ketosis and milk production in dairy cows in early lactation. To detect ketosis [blood ß-hydroxybutyrate (BHB) ≥1.2 mmol/L], Holstein cows were screened daily from 3 to 16 d in milk using a cow-side meter. Cows diagnosed with ketosis (n = 104) were randomly assigned to twice-daily milking (TDM) or reduced to once-daily milking (ODM) for 2 wk, then returned to twice-daily milking. Both treatment groups received a 5-d treatment of an oral propylene glycol drench (PG; 300 g) beginning on the afternoon of the diagnosis; cows received additional 5-d PG treatments if they had a ketotic test result (blood BHB ≥1.2 mmol/L) at least 4 d after finishing the first PG treatment. Blood BHB tests were conducted for the first 3 d after ketosis diagnosis, and then once every 3 d for 21 d of trial (DOT). Milk and milk component data were collected weekly for 15 wk following trial enrollment. The ODM group showed rapidly and markedly decreased blood BHB concentrations (primiparous cows: 1 DOT, 0.92 ODM vs. 1.22 TDM, 15 DOT, 0.55 vs. 0.81 mmol/L; multiparous cows: 1 DOT, 1.01 vs. 1.40, 15 DOT, 0.78 vs. 1.65 mmol/L). In addition, a logistic regression model indicated that ODM cows were less likely to have blood BHB concentrations ≥1.2 mmol/L [primiparous cows: 3 DOT: ODM 1% (95% confidence interval: 0-10%) vs. TDM 43% (30-58%), 15 DOT ODM 0% (0-0.2%) vs. TDM 22% (13-36%); multiparous cows: 3 DOT: ODM 33% (24-44%) vs. TDM 59% (48-69%), 15 DOT ODM 20.9% (13-31%) vs. TDM 64% (53-74%)]. The proportion of ODM cows that required additional treatments of PG were substantially lower than the TDM group (ODM: 39%; TDM: 64%) than the TDM cows during the initial 21-d period. However, during the 2-wk treatment period, cows in the ODM group produced 26% less milk and 25% less energy-corrected milk than the TDM cows. During wk 3 to 15, when all cows were milked twice daily, ODM cows produced less milk (-14%) and energy-corrected milk (-12%) compared with the TDM group. Milk protein percentage was greater, and milk fat percentage and linear score tended to be greater in the ODM group over 15 wk. In conclusion, a 2-wk reduction of MF in ketotic cows from twice to once daily with treatment with PG resolved ketosis and decreased blood BHB concentrations more effectively than treating TDM cows with PG alone. However, the 2-wk MF reduction had immediate and long-term (up to 13 wk after cessation of MF reduction) negative effects on milk production.

Impaired locomotion (lameness) may negatively affect the ability and desire of cows to milk voluntarily, which is a key factor in success of automated milking systems (AMS). The objective of this study was to identify factors associated with herd-level lameness prevalence and associations of lameness and other farm-level factors with milking activity, milk yield, and milk quality in herds with AMS. From April to September 2019, 75 herds with AMS in Ontario, Canada, were visited, and data on barn design and farm management practices were collected. Data from AMS were collected, along with milk recording data, for the 6-mo period before farm visits. Farms averaged 98 ± 71 lactating cows, 2.3 ± 1.5 robot units/farm, 43.6 ± 9.4 cows/robot, 36.4 ± 4.9 kg/d of milk, a milking frequency of 3.01 ± 0.33 milkings/d, and a herd average geometric mean SCC of 179.3 ± 74.6 (× 1,000) cells/mL. Thirty percent of cows/farm (minimum of 30 cows/farm) were scored for body condition (1 = underconditioned to 5 = over conditioned) and locomotion (1 = sound to 5 = lame; clinically lame ≥3 out of 5 = 28.3 ± 11.7%, and severely lame ≥4 out of 5 = 3.0 ± 3.2%). Clinical lameness (locomotion score ≥3) was less prevalent on farms with sand bedding, with increased feed bunk space per cow, and on farms with non-Holstein breeds versus Holsteins, and tended to be less prevalent with lesser proportion of underconditioned cows (with body condition score ≤2.5). Severe lameness occurrence (farms with any cows with locomotion score ≥4) was associated with a greater proportion of underconditioned cows and in farms with stalls with greater curb heights. Herd average milk yield/cow per day increased with lesser prevalence of clinical lameness (each 10-percentage-point decrease in clinical lameness prevalence was associated with 2.0 kg/cow per day greater milk yield) and greater milking visit frequency per day, and tended to be greater with increased feed push-up frequency. Lesser herd average somatic cell count was associated with lesser clinical lameness prevalence, herd average days in milk, and proportion of overconditioned cows, and somatic cell count tended to be lesser for farms with sand bedding versus those with organic bedding substrates. The results highlight the importance of minimizing lameness prevalence, using of sand bedding, ensuring adequate feed access and feed bunk space, and maintaining proper cow body condition to optimize herd-level productivity and milk quality in AMS herds.
